Research Abstract

Streptococcus intermedius has increasingly been isolated from oral and liver abscesses. We had found a carbohydrate antigen CD15s-related on this bacterium. CD 15s is a ligand for selectins, and is expressed on hepatocytes related to necroinflamrnatory changes. The aim of this study was to determine the effect of CD15s-related antigen on liver diseases. We immunized BALB/c mice from gingiva with a large number of killed S. intermedius, then anti-CD 15s antibody titers were measured. Tissue sections of the liver were observed immunohistochemically, and also stained by TUNEL method for detection of apoptotic cells. Lymphocytes from the liver were analyzed by flow cytometry. As the result of immunization, an increase in the titer of anti-CD15s serum IgG was observed. In the liver, inflammation occurred in 31.6% of the immunized mice, and hepatocytes around the inflammatory lesion expressed CD15s cytoplasmically. Apoptosis of the hepatocytes in and around inflammatory lesion was demonstrated. Most of the infiltrating T cells in the liver were CD8ィイD1+ィエD1T cells. Our results suggest that immunization with CD 15s-related positive S. intermedius from gingiva induced the systemic immune response and trigger autoimmune response in the liver.
