Research Abstract

The aim of this study was to investigate on three dimensional bone formations on the surface of several biomaterials using osteoblastic primary culture. However, because of several restrictions regarding osteoblastic primary culture, including fetal bovine bone cells, I've just found that osteoblastic primary culture derived from embryonic porcine mandibular bone shows matrix calcifications under some circumstances so far.
When commercially available osteoblastic primary culture derived from human bone was cultured with hydroxyl apatite granules, it showed interesting structures that was not seen in the two dimensional cultures. This means osteoblastic primary cultures may be powerful tools to study the mechanisms of three dimensional bone formations in vivo when co-cultured with biomaterial granules. However, the biggest problem was that proliferation and differentiation of those cells were very slow and the viability of those was too low.
Now, a new osteoblastic primary culture derived from embryonic porcine mandibular bone was developed. This culture system showed rapid calcification among surrounding matrix under some circumstances. Elucidation of the optimal conditions for rapid calcification of matrix still needs to be done.
