Research Abstract

多剤耐性遺伝子MDR1の発現制御には転写因子YB1(Yboxbinding protein)の関与を認めてきた。紫外線照射により、細胞質に存在するYB1は核に移行しMDR1遺伝子や他の遺伝子の転写を制御していると考えられている。本研究では、1.YB1に結合する蛋白IRP(iron responsive element binding protein)を酵母Two-hybrid法を用いて単離した。Pull down assay によりin vitroにおいても結合を確認した。この蛋白は、翻訳の調節に関与しており、抗がん剤耐性において、発現の増大が認められた。
2.耐性関連遺伝子YB-1とp53との結合をin vivo,in vitroにおいて確認した。P53はMDR1遺伝子の発現を抑制しており、MDR1プロモーター上のYboxを介して、調節していることを明らかにした。YB1がストレスに応じて種々の遺伝子の発現調節に関与する事が示唆された。
3.肺癌においてYB1の核移行の有無と予後に有為な相関が認められた。
