アデノ随伴ウイルスのRep蛋白質の変異導入による機能解析

Research Abstract

昨年度に実施した、アデノ随伴ウイルス(AAV)のRep蛋白質の変異導入による解析により、細胞毒性が減弱しているEK57AA、RD61AA、R122A、R185A、E201Aの計5クローンを同定した。これら5クローンが本当に野生型Rep蛋白質と同程度のAAVS1特異的組込み活性を保持しているか詳細に調べるため、AAVのinverted terminal repeat(ITR)配列ではさんだNeo遺伝子プラミスドと変異Rep蛋白質発現プラミスドを293細胞にトランスフェクションし、G418存在下で培養を行った。そこから得られたクローンをランダムに拾いそれらのサザン解析を行い、Neo遺伝子がAAVS1領域に組込まれている頻度を検討した。野生型Repを用いた場合は60%(12/20)のクローンでAAVS1特異的組込みが認められ、E201Aを用いた場合、89%(25/28)のクローンでAAVS1特異的組込みが確認できた。一方、その他のEK57AA、RD61AA、R122A、R185Aを用いた場合はAAVS1特異的組込みを示したクローンの頻度は20%以下であった。以上のことは、E201Aが野生型Repと同程度、もしくは、より高い部位特異的活性を保持していることを示しており、細胞毒性が減弱していることと考え合わせ、染色体の特定の部位に任意の外来性遺伝子を導入するテクノロジーにより適している可能性があると考えられる。
