Research Abstract

本研究では、前年度に引き続き、(1)実際に各種白血病患者から腫瘍細胞を分離し、細胞内TNF発現量とanthracyclin系抗癌剤(DOX)に対する感受性との相関を調べた。その結果、DOX感受性と細胞内TNF発現量との間には、強い逆相関関係が認められた。(2)各種白血病細胞株における、細胞内TNF発現量と、Inhibitor of apoptosis(IAP)familyであるsurvivinの発現量との関係について調べた結果、正の相関傾向がみられた。そこで、構成的に細胞内TNFおよびsurvivinが発現しているHL-60細胞を、all-trans-retinoic acid(ATRA)で処理後、両者の発現を5日目まで経時的に調べた。その結果、両者の発現量は分化誘導にともない明らかに低下した。(3)抗癌剤治療の前後で細胞内TNF発現量を測定し、anthracyclin系抗癌剤、あるいは他の抗癌剤の治療効果予測マーカーとして臨床応用が可能か否か検討した。Anthracyclin系抗癌剤で治療された患者群で、治療に抵抗性を示したno response群では、治療が奏功したpartial response及びcomplete response群に比べ、治療前の平均細胞内TNF発現量が約2倍高かった。前年度の実績および本年度の実験結果から、細胞内TNFの、白血病における自然耐性および獲得耐性因子としての意義が明らかとなり、抗癌剤に対する新しい治療効果予測マーカーとしての有用性が示唆された。
