Research Abstract

1)一過性脳虚血後の内因性アポEの動態
一過性前脳虚血後の海馬遅発性細胞死においては細胞死が明らかになる前から神経細胞にアポEが沈着し始めることが明かとなった。この間にアストロサイトに見られていたアポEは減少し、同時にアポE受容体が神経細胞に強く発現していた。このことより神経細胞は受容体を介しアポEを細胞内に取り込んでおり、これを介し治療薬を細胞内に投与することが可能と考えられた。
2)アポEcDNAのクローニング
ラット脳及び肝臓から、アポEcDNAのクローニングを行ったところ、脳のアポEは一般に見られる肝臓のそれと比べ3塩基異なるイソ蛋白であることが分かった。これを大腸菌に発現させGST-fusion protienとして精製した。
3)神経細胞への影響
神経細胞の初代倍養系にこの蛋白を投与したところ。細胞内への取り込みがみられた。過剰量では毒性も見られたが、適量により若干の保護効果が見られた。現在この蛋白を用いカルシウムキレート剤を封入し保護効果の所見が得られつつある。
