Research Abstract

当教室では,子宮内膜症患者腹水中には高濃度のTumor necrosis factor α(TNFα)とInterleukin-8(IL-8)が優位な相関をもって存在すること,赤色の子宮内膜症活動性病変の拡がりがTNFαおよびIL-8濃度とも相関することを報告した.TNFαはサイトカイン誘導能を有することが知られている.子宮内膜症由来間質細胞におけるTNFαのIL-8誘導能を検索し,TNFαの細胞増殖及ぼす影響を検討した.患者の同位のもと,手術時に採取した卵巣チョコレート嚢胞壁(n=13)から間質細胞をOsteenらの方法を用いて分離培養し実験に供した。RT-PCRおよびSouthern blot法により内膜症由来間質細胞においてTNFα受容体typeIおよびtypeII遺伝子の発現を確認した.免疫染色により,内膜症間質細胞でのIL-8蛋白の発現が観察された.Northern blot法により0-100pg/mlのTNFαの添加はIL-8遺伝子の発現を増強し,培養上清中のIL-8濃度を添加濃度および時間依存性に増加させた.TNFα(20-100pg/ml)は内膜症間質細胞の(122%)増殖を有意に促進し,その促進作用は抗TNFα抗体のみならず抗IL-8抗体の併用添加でも中和された.
以上の成績から子宮内膜症由来間質細胞においてTNFαはIL-8の遺伝子発現と産生を誘導した.IL-8を介するTNFαの間質細胞増殖促進作用は子宮内膜症の進展に関与する可能性はじめて明らかにした.
