Research Abstract

体重減少性無月経患者では夜間血中メラトニン濃度が高値を示し、その増量が本症のLHパルス発現障害に関与していることを明らかにしてきた。
本年度は、ラットFood restriction(FR)モデルを用い、体重減少がメラトニン産生動態に及ぼす影響を松果体中メラトニン産生関連代謝物、メラトニン産生酵素活性、血中メラトニン濃度、メラトニン産生を刺激するNorepinephrine(NE)に注目し検討した。Sprague-Dawley雌ラットを用い14h明、10h暗の条件下で飼育し、自由摂食させた群(AL群)と排卵周期確立後の8週0日から摂食量を50%に制限したFR群に群別し、10週0日のMid darkに体重、子宮、卵巣重量を測定し、松果体および血液を採取した。松果体中メラトニン、Tryptophan(T)、Serotonin(S)、NAS、M産生酵素活性(NAT,HlOMT)、NE含量および血中M濃度をHPLC法で測定した。性周期は膣スメアで確認した。その結果、1)FR群の体重、子宮、卵巣重量は、AL群に比し有意に減少した(各々22.4、41.7、36.4%減少)。2)FR群の4days cycleの性周期は、8週6日から9週4日の間に全て消失した。3)松果体中メラトニン、メラトニン産生前駆物質(T,S,NAS)の含量、メラトニン産生酵素(NAT,HIOMT)活性、およびNE含量については、両群間に差を認めなかった。一方、4)血中メラトニン濃度は、AL群(59.99±23.13pg/ml)に比較しFR群(92.50±26.39pg/ml)が、有意に高値であった。以上よりFRに伴う血中メラトニン濃度の増量が示された。更にこのメラトニンの増量は松果体でのメラトニン産生能の増加に基づかず、体重減少に起因するメラトニンの末梢での代謝要因が関与している可能性が強く示唆された。
