Research Abstract

EGCGの胆汁中の代謝物の検索に加えて,今年度は薬物が吸収される前段階,すなわち消化管内におけるEGCGの変化を検討するために,腸内細菌によるEGCGの代謝物の検索を行った.EGCGをラットから得た腸内細菌と24時間インキュベーションを行った結果,HPLC分析においてEGCGの分解物と思われるピークを数種認めたので,カラムクロマトによる分離,精製を行い,methylgallateを単離した.その他の分解物については現在分離,精製中である.
また,胆汁中におけるEGCGの安定性についても検討を行った.その結果,EGCGをラット胆汁中でインキュベーションを行い,EGCGの二量体に相当するtheasinensin Aの生成を認めた.
以上,本課題研究において,経口投与後,代謝物として胆汁中にEGCGおよび微量ながらそのジメチル体を確認した.また,腸内細菌によるEGCGの代謝の検討を行い,methylgallateを単離,さらには胆汁中でEGCGは一部theasinensin Aに変換することも見出した.
これらの結果はEGCGが経口投与された後,初回通過効果を考慮に入れてもEGCG又はその代謝物が循環血に入っていること,またEGCGが腸内細菌又は胆汁中で変化を受けてから吸収される可能性を示唆するものである.今後,EGCGのさらなる代謝物の検索およびそれら代謝物とEGCGの活性の比較検討等を行い,EGCGの多様な生理活性の作用部位,作用機序を究明し,化学発癌予防に貢献し得るデータを蓄積する必要がある.
