Research Abstract

(1)摘出血管組織・単離細胞を用いて、NO関連物質による血管弛緩反応におけるマキシK^+チャネル活性化の寄与を検討した。
(2)NOドナー(ニトログリセリン・NOR3)や心房性ナトリウム利尿ホルモン(ANP)によるラット大動脈および腸間膜動脈の弛緩反応を検討したところ、腸間膜動脈の弛緩反応はイベリオトキシンで著明に抑制されるものの、大動脈の反応は影響されないことを見出した。
(3)ウェスタンブロット法による蛋白定量・免疫組織染色法を用いて、ラットの大動脈および腸間膜動脈平滑筋に発現しているマキシK^+チャネルレベルを測定したところ、両者の間に有意な差は認められなかった。
(4)単離平滑筋細胞を用いて、パッチクランプ法によりマキシK^+チャネル電流を測定したところ、大動脈平滑筋細胞においてもマキシK^+チャネル電流が認められた。
(5)大動脈標本のNOドナーによる弛緩反応はBa^<2+>により強力に抑制されることから、主として内向き整流性K^+チャネルの活性化が関与することが示唆された。
(6)腸間膜動脈のNOドナーによる弛緩は、タプシガルギン処置により顕著に抑制されることから、細胞内Ca^<2+>ストア機能とマキシK^+チャネル活性化の機能的カップリングの可能性が示唆された。
(7)以上の結果から、NOドナーによる血管弛緩は、抵抗性動脈ではマキシK^+チャネルの活性化が、導管動脈では内向き整流性K^+チャネルの活性化が関与することが示唆された。マキシK^+チャネルが大動脈平滑筋細胞においても発現レベルが腸間膜動脈平滑筋細胞と差が無いにも拘わらず、NOドナーによる弛緩に寄与しない理由は現在のところ不明である。今後、抑制性β-サブユニットによる修飾効果をはじめとする調節機序を検討する予定である。
