Research Abstract

RNAポリメラーゼIII(Pol-III)は、5S rRNA、tRNAや一群のsnRNAを合成する重要な転写装置である。しかし、植物におけるPol-IIIの転写機構の研究は皆無と言ってよい。
そこで、タバコBY-2培養細胞の核より再現性のよい高活性のPol-III依存in vitro転写系を確立した。
本年度は、このPol-III依存in vitro転写系に、イントロンを持つ遺伝子からのtRNA前駆体のスプライシング活性も有することを明らかにしたので、反応系の改良を進め、活性の高い系の確立に成功した。転写とスプライシングはマグネシウム濃度を変化させることにより人為的にコントロールすることを見出したので、各々を個別にあるいは連動した系として反応の解析が可能となった。低濃度のマグネシウム(1mM)ではtRNA前駆体のみが蓄積されるので、これにより正確な転写開始と停止部位を決定できることになった。またスプライシング部位もプライマー伸長法を用いることにより正確に同定できることを示した。
