毛状根を利用したテンサイの遺伝的相補性検定システムの確立

Research Abstract

(1)ビートシストネマトーダ(Heterodera schachtti)に対して感受性を示すテンサイ品種TK81-0と抵抗性の野生種Beta procumbensの芽生よりリーフディスクを調製した。リーフディスクにAgrobacterium rhizogenesを接種し不定根の誘導を企てた。その結果、両遺伝子型において低頻度ながら不定根が分化し、さらに不定根の培養を通じて毛状根を得た。
(2)カルスを経由するトランスフォーメーション系の開発を企てた。遺伝的に多様な20品種を供試して幼胚軸由来のカルス培養を試みた。その結果NK219-0とNK219-MSという2品種がカルス形成率、不定芽(又は不定胚)形成率共に極めて高い値を示した。
(3)Agrobacterium tumefaciensを感染させたカルスの中には、共存培養中にネクロシスを起し枯死するものも少なからず認められた。しかし、カルス形成培地上での継代培養期間を短縮することによってネクロシスの発生をかなり回避することができた。培養期間が6週間以内のAgrobacterium感染カルスより不定芽(まれに不定胚)の形成がみられた。不定芽(又は不定胚)形成までに要する期間は40日程であった。GUSアッセイを行ったところ、GUS遺伝子の発現を示す青色スポットが明瞭に見出され、テンサイ形質転換系として今後活用し得ると期待される。
