Research Abstract

生殖器官の凍結保存を実現するために、薄切組織を遊離細胞と同様に急速冷却する手法、臓器摘出に際して動脈から耐凍剤を灌流した後薄切して凍結する手法など、様々な試みが行われてきたが、受精能をもつ卵子をほとんど得られていない。遊離細胞と比較して巨大な器官を凍結保存するためには、ブレークスルー的技術革新が必要である。本研究は、極地に生息する動物の体液の氷点を低下させ、氷晶の成長を抑えて氷晶による細胞破砕を阻止する耐凍蛋白遺伝子、アポトーシス阻止因子遺伝子などを生殖器官にex vivo導入して凍害保護と細胞死抑制することで生殖器官の凍結保存を実現しようとした。遺伝子導入のためリポソーム作製法う確立し、生殖器官に導入した遺伝子を特異的に発現させるex vivo遺伝子導入法の開発を進めた。遺伝情報の伝達を担う生殖細胞に外来遺伝子が不用意に導入されることを未然に防がなくてはならないので、導入効率は高いがウイルス遺伝子を含むウイルスベクター法はとらず、ウイルス遺伝子などの混在遺伝子を含まない「リポソーム法」を開発した。すなわち、リン脂質とコレステロールからなるリポソーム膜を作製し、卵胞特異的に遺伝子を導入させるべく、標識となる卵胞特異的糖蛋白を癒合させた。さらに、細胞膜融合活性をもつエンベロープ蛋白を紫外線にて不活化したセンダイウイルス(HVJ:hemaggulutinating vieus of Japan)から調製し、リポソーム膜と癒合させた。このように調製した「HVJ-リポソーム」と標的細胞が融合してリポソーム内の遺伝子が効率よく導入・発現する系を確立した。次いで、卵胞細胞特異的レクチンをリポソームに組込んで遺伝子の組織特異的組込率を高め、high mobility group |を標的遺伝子の上流に組込んでと安定発現を計った。
