Research Abstract

ゼノパス(カエル)形態形成(変態)に、免疫系によるトリミングが関与することを初めて明らかにした。我々は既に、同系カエルの幼生皮膚が成体免疫系によって拒絶されることを報告している。本プロジェクトでは、アフリカツメガエルの幼生の尾部が消失する過程で、単核球が浸潤すること、生体のT細胞が幼生抗原を認識し、増殖反応を生じ、その後キラー細胞となること、その結果尾部の上皮細胞がアポトーシスになることをin vitroの系で明らかにした。成体T細胞の幼生抗原認識は、直接幼生上皮の抗原を認識する系路、いったん成体抗原提示細胞内で幼生抗原が処理され、それが抗原提示細胞膜のMHCクラスII分子上に提示される系路の二種があることを明らかにした。
幼生皮膚を成体に複数回移植し、抗幼生抗原特異的抗血清を得た。この抗血清を用い、分子量53、59kDaの蛋白を同定した。尾部の幼生抗原は変態期直前をピークに、尾消失まで発現するが、成体上皮細胞では変態期以降消失することを明らかにした。さらに、幼生特異蛋白のアミノ酸配列をもとに、幼生抗原の遺伝子断片を同定し、それに基づいて作製したペプチド分子に対して成体T細胞が反応することを確認した。
現在、幼生抗原全分子、全遺伝子の同定を行いつつある。
