老化と血栓症:老化モデルマウスでは血液凝固線溶因子の組織内発現異常が存在するか?

Research Abstract

昨年度の研究において我々は、エンドトキシン(Lipopolysaccharide:LPS)5μgを腹腔内投与した若年マウス(C57BL6/J系統オスの8週齢)に比較し、同様にLPSを投与した老齢マウス(12カ月齢および24カ月齢)において、血中の活性型PAI-1(Plasminogen activator inhibitor-1:主要な線溶制御因子)抗原量と、肝、腎などの組織におけるPAI-1mRNA発現量のより顕著な増加を観察したと報告した。今回、このPAI-1発現の増加と組織における血栓傾向との間にどのような関連性があるかについて検討するため、LPS投与後の腎糸球体の微小血管内におけるフィブリン沈着量を半定量的に解析し、若年、老齢マウス間での違いについて比較検討を行った。LPS5μgをそれぞれのマウス(若年、老齢5匹ずつ)に腹腔内投与し、2,4,8,16,24時間後に腎(生理食塩水にて灌流後のもの)を摘出して、抗マウス・フィブリン抗体を用いた免疫組織化学法によってフィブリンを染色し、その沈着を認める腎糸球体数をカウントした。その結果、老齢マウスのほうがより高度なフィブリン沈着を認め、特にLPS投与4時間後にはフィブリン陽性の糸球体の割合が、若年マウスで17%に対して老齢マウスでは57%、8時間後には若年マウスでわずか3%に対して老齢マウスでは42%と、顕著な差を呈していた。このフィブリン沈着量の時間推移は、老齢マウスの肝、腎におけるPAI-1発現増加の時間経過とよく相関しており、実際の血栓形成におけるPAI-1発現の重要性と、敗血症という病態において老齢マウスでは血栓傾向が著明に増大していることが示された。
