Research Abstract

リボソームはmRNAの塩基配列に従ってポリペプチド鎖を正確に伸長し,タンパク質を合成する細胞内小器官である.しばしば,蛋白質合成工場に例えられる通り,遺伝暗号解読,ペプチジルトランスファー,トランスロケーション,分子的な校正など,生命にとって極めて重要でかつ複雑な仕事を整然と行なっており,その構造が決定された場合の生物科学全体に与えるインパクトは計り知れないものがある.本研究では,リボソームの原子レベルでの立体構造解析を行なうための,高分解能のX線回折を与える結晶を調製することを目的とした.このため,好気性の超好熱古細菌Aeropyrum pernix(至適生育温度90度)嫌気性の超好熱古細菌Pyrococcus horikoshii(至適生育温度98度),高度好熱真性細菌Rhodothermus obamensis,からリボソームを調製することを試みた.超遠心分離法,疎水性クロマトグラフィー,ショ糖連続密度勾配遠心法を用いて,70Sリボソーム,各サブユニットの調製方法を確立した.また,超遠心分離で粗精製した70Sリボソームは,さらに疎水クロマトグラフィーを用いることで,より純度高く精製することができることを確認した.すべてのリボソームサンプルについて,蛋白質合成活性を調べ,高温(70度)において活性があることを確認した.今後は,さらに結晶化条件の検討を行いX線構造解析に持ち込むことを計画している.
