Research Abstract

FK506結合蛋白質(FKBP)は免疫抑制剤FK506と結合するとカルシニューリンの機能を阻害し、免疫担当細胞の働きを抑えることが知られている。我々はテトラヒメナにもFKBPが存在することを発見した。これは、この蛋白質が普遍的な生理機能に関与することを示唆している。この普遍的な生理機能を明かにすることが本研究の目的である。
1.テトラヒメナのFKBP遺伝子をクローニングし、そのcDNAの全塩基配列を決定した。推定されたアミノ酸配列からテトラヒメナのFKBPは分子量12kDaでFK506結合部位を保持していた。ホモロジー検索の結果、ショウジョウバエ、ヒト、マウスのFKBP12と約60%の相同性を持ち、類似性は80%近いことが判った。
2.テトラヒメナの同調分裂細胞をFK506で処理したところ細胞質分裂が阻害されることが明らかになった。
3.テトラヒメナFKBP12の機能を明らかにするため:(1)抗-FKBP12抗体を用いたFKBP12の細胞内局在性の決定、(2)FKBP12に結合するカルシニューリンのような蛋白質の同定、などを計画した。その第一段階として大腸菌でリコンビナントFKBP12の大量発現を行い、リコンビナントFKBP12をウサギとモルモットに注射して抗-FKBP12抗体の作製に成功した。
