Research Abstract

糖鎖は細胞の表面の大部分を覆っており、細胞間や細胞一基質間相互作用に重要な役割を果たしていると考えられている。われわれは糖蛋白質糖鎖の系統だった解析法をほぼ自動化することに成功し、その機能を解明しようと試みている。ヒト脳の糖鎖解析を行ったところ、脳の各部位においてN-結合型糖鎖発現パターンに個人差はほとんどないことが明らかとなった。本年度は老化と密接に関連したアルツハイマー病患者の脳内糖蛋白質糖鎖の解析を行った。10%ホルマリン固定ヒト病理標本、前頭葉皮質および前頭葉白質をアセトン沈殿し解析に用いた。蛍光標識後にノイラミニダーゼ処理し、シアル酸を除去した後に糖鎖構造解析のための2次元HPLCを行った。その結果、正常人と糖鎖パターンに差のあることが見いだされた。今後、これがどのような糖鎖構造を反映しているのか解析する。
