Research Abstract

バベシア症は、ダニによって媒介されるバベシアが宿主動物の赤血球に寄生して、発熱、貧血、黄疸等の症状を呈する原虫感染症であり、家畜や犬などで世界的に認められておりで毎年数兆円にのぼる経済的被害をうけている。本研究では、家畜バベシア症に対する新規の迅速・簡便遺伝子診断法の開発と新規治療薬の検索を目的に、以下の項目について検討した。
1)新規の遺伝子の検索と診断法および予防法への応用研究
これまで、B. bovisの微細構造であるスフェリカルボディー4の組換えBbSBP-4蛋白質(BbSBP-4)を用いたELISAは最も高い特異性と感度を有することを明らかにした。また、牛バベシアB. bigemina AMA-1遺伝子を特異的に検出するnested PCRを開発した。
24年度は、馬バベシアB. caballiのロプトリー蛋白質を効率よく産生する方法を開発した。これらの方法を用いて、モンゴルの牛および馬のバベシア症に関する分子疫学的調査を行った。その結果、B. bigemina, T. equi, B. caballiがモンゴルにおいて高率に感染し、モンゴルに特有な遺伝子配列が存在することが明らかとなった。
2)アピコプラスト等をターゲットにした新規治療薬の検索
これまでバベシア原虫のアピコプラストが薬剤標的として有望であることを、遺伝子解析、DNAの転写阻害剤、アピコプラストの重要な酵素であるペプチド脱ホルミル酵素の阻害剤を用いた実験により、明らかにしてきた。24年度は、牛バベシアBabesia bigeminaヒートショック蛋白質(BbigHSP-70)の遺伝子解析を行った。更に組み替え抗原を作製し、これに対する抗血清がB. bigemina, B. bovis, B. caballi, B. gibsoni,とB. microtiの溶解抗原と反応することが示された。更に、BbigHSP-70で免疫したマウスは非免疫群に比較して、B.microtiの攻撃感染に対し強い免疫効果を示すことが明らかにされた。さらに、Babesia bovisのヒストン脱アセチル化酵素(HDAC)の分子生物学的解析を行い、その阻害剤であるアピシジンが培養バベシアの増殖抑制を示した。今後、これらはバベシア症に対する新たな薬剤として更に検討にすることが期待される。
