Research Abstract

1.Candida albicansを特異的に検出するデザインペプチドの設計
昨年度の研究において、C. albicansの病原性プロテアーゼであるSapをターゲットとして、C. albicansを特異的に攻撃可能なデザインペプチドの設計が完了していた。
本年度では、病原性プロテアーゼによって特異的に切断されるリンカー配列に蛍光基と消光基を結合させることで、カンジダ症診断薬のプロトタイプを作成した。この消光ペプチドを、クエン酸バッファー中でSapと混ぜ合わせたところ、20分ほどで強い蛍光が観察され、短時間でカンジダ症を診断できる可能性を示唆した。また、実際の臨床の条件を模するために、模擬唾液サリベートを用いて実験を行ったところ、その場合でも強い蛍光が短時間で見られることがわかった。
2.C. albicansのプロテオーム解析と新規病原性因子の同定
C. albicansが血中に侵入すると、致死率が非常に高い全身性カンジダ症を発症するが、その機構はよくわかっていない。そこで、未知病原性因子を網羅的に決定するために、プロテオーム解析を行った。具体的には、C. albicansをfetal bovine serum中で培養し、その条件において特異的に発現が上昇するタンパク質群をLC-MS/MSによって決定したところ、数十個の因子が見つかった。その中でも、我々がBlplと名付けたタンパク質は12倍もの発現上昇率を示したため、Saccharomyces cerevisiaeの異種発現系を用いて、その機能を調べた。その結果、Blplは各種ストレス耐性に関わる因子であり、熱耐性・抗生物質耐性・塩濃度耐性などを上昇させた。
