Research Abstract

オオムギは穂の形態から二条性と六条性に大別される.二条穂は主列小穂のみ穀粒をつける.一方,六条穂は側列小穂も稔実するため二条穂に比べて3倍量の穀粒をつける.条性はHD-Zip I型転写因子をコードするVrs1遺伝子に制御されることが知られている.しかし,VRS1タンパク質の分子機能や発現パターンは十分に解明されていない.また,近縁種のコムギ,イネ,トウモロコシなどにおけるVrs1相同遺伝子の存在や機能はほとんど知られていない,そこで,本研究ではオオムギでの詳細な発現解析とタンパク質機能解析,そしてイネ科での比較ゲノミクス解析を行なった.オオムギにはVrs1のパラログとしてHvHox2が存在し,HvHox2オーソログがイネ科で高度に保存されていることがわかった.HvHox2は幼穂,葉,根など全身で発現しているのに対してVrs1は幼穂だけで発現していた.Vrs1は遺伝子重複によって生じ,機能分化したということが示唆された.機能解析を進めた結果,HvHOX2とVRS1タンパク質はC末端領域に保存された転写活性化ドメインを持つことがわかった.HvHox2は他のHD-Zip遺伝子と同様,維管束に局在していた.一方,Vrs1は側列小花で特異的に発現し,特に雌蕊に局在していた.Vrs1遺伝子とは別座の六条変異体(vrsw, vrs3, vrs4, int-c)の発現解析によって,Vrs1の転写がVrs4によって促進されていることが明らかになった,以上の結果からVrs1遺伝子は遺伝子重複後に新しい発現パターンを獲得することによって機能分化し,側列小花の発達を抑制していることが明らかになった.
