培養神経系における情報処理機能の人為的制御と再生医療への応用

Research Abstract

最終年度においても引き続き微小流路内におけるラット大脳皮質神経細胞のパターニング培養法の開発と改良を主に行い、パターニング培養神経回路の形成とその機能制御について解析を行った。特にこれまでの実験で不十分であった,形成神経回路網のシナプス極性や軸索伸展方向の定量解析のための免疫染色法・活動計測のためのカルシウムイメージング法を重点的に行った。また、マイクロウェル部のアレイ構造と化学パターニング構造のデザインを変更することで,多種多様な構造と機能を有した培養神経回路の形成制御を試みた。これらの結果はJournal of Bioscience and Bioengineering雑誌に投稿し、受理・掲載されている。また研究課題に取り組む中で得られた微細加工技術・その培養細胞への応用法を、研究課題の中心となる大脳皮質神経細胞のみでなく、他の細胞種(C2Cl2筋芽細胞等)に応用することで様々な培養組織の形態・機能制御についても検討を行い、これらの実験についても外部発表の準備を行っている。これ以外にも該当研究者の有する実験技術・知見を研究室内外の研究者と協力することで多数の研究業績を発表するに至っている。本研究で提案した実験手法の今後の発展により、回路網構造・情報伝達経路を人為的・任意に構築した培養神経回路網を形成し、その情報処理機能を制御・応用することが可能となると考えている。また、その先には構築した実験系の特徴を用いて、脳神経系のある局所神経回路構造の形態・機能を再現した人工培養神経チップによる損傷脳機能の修復・代替法の開発という目標を持って今後の研究進展を行いたい。
