Research Abstract

環状ゲノムを持つバクテリアにおいて,様々な要素がそのゲノム上に対称あるいは非対称的に存在しており,そのゲノム構造を維持している.中でも最も特徴的なゲノム対称構造は複製開始点と終結点の関係であるが,全てのバクテリアゲノムにおいてこの対称構造が観察される訳ではない.環状ゲノムを持つバクテリアにはリーディング鎖とラギング鎖におけるグアニン(G)とシトシン(C)の偏りを,(C-G)/(C+G)で表現したGCskewがほとんどの種において対称的な傾向が認められ,現在の複製開始・終結点の決定にはこの塩基組成の偏りに基づいたものが主流になっている.ところが,塩基組成だけではバクテリアゲノムで多く起こっている大規模な変異には対応できない.そこでまず,バクテリアの複製終結点として働いていると考えられるようになったdif配列を基にした新たな予測手法を確立する事を目指した.以上の事から,これまで我々は複製終結点の高精度な予測のためのdif配列予測手法を確立し,ゲノムワイドに見られる対称性を維持するシステムの解明を目的とした研究を行ってきた.その結果641種・714染色体でのdif配列を予測する事に成功し,dif配列と塩基組成バイアスはほぼ同じ場所に位置している事が判明した(Kono et a1.2011).さらに,複製機構のモデルを計算機的に構築し,バクテリアが長い進化の歴史の中でどのような複製機構を使ってきたのかをシミュレーションし,ゲノム構造が構築されてきた進化の流れを議論する事が出来た(Kono et a1.2012).そして本研究では,これまでの計算機的知見を元に,in vivoで複製開始・終結点の位置を観察し,複製終結の配列とタンパクの関係を解き明かす実験的なアプローチを行った.その結果,生体内での実際の複製終結点の位置を実験的に証明する事に成功した.
