Research Abstract

1.プロウログアニリンにおける構造形成機構の解明
今までに、プロウログアニリンの構造形成機構の解析により,プロウログアニリンの構造形成はウログアニリン領域内ジスルフィド結合の架け違った異性体1および異性体2を経由して天然型のジスルフィド結合様式を形成することを明らかにしている。さらにそれぞれの異性体の構造決定を目指し,NMR解析に着手した結果,最終的にトラップされる異性体2はプロ領域の構造変化を駆動力として生理活性構造の構築を行っていることを強く示唆した。現在,構造決定の解析中であり,本成果の達成はプロウログアニリンのフォールディングにおける原子レベルでの解明に繋がる。
2.機能性人工タンパク質の創製技術の展開
プロ領域の分子内シャペロン機能を構造制御されるペプチドホルモンの立場から解析するため,今までにプロ領域に捕らえられる人工ペプチドの創製に成功している。そこで,本年度はさらにその構造形成機構を詳細に検討した結果、正しいジスルフィド結合を持っているが不活性構造(トポロジカル異性体)を経由して活性構造へフォールディングすることを明らかにし,プロウログアニリンにおけるプロ領域の構造制御機構と異なった構造制御が行われていることを明らかにした。
さらに以上研究成果を通じて,新たなフォールディング促進剤を創製し論文に採用された結果,FEBS Jにおいて該当分野の総説を、国内外の他の研究グループを招き、まとめるに至った。
