植物におけるsmall RNA依存的翻訳抑制機構の生化学的解析

Research Abstract

RNAサイレンシングは、多くの真核生物に共通して存在し、20数塩基のsmall RNAを介した塩基配列特異的なRNA切断、翻訳抑制、あるいは転写抑制による遺伝子発現制御機構として機能する。動物では翻訳抑制及び訳NAの不安定化によって遺伝が制御されているが、植物では切断による制御が一般的であると言われていた。しかし最近になって植物でもsmall RNAを介した翻訳抑制が広く行われていることが明らかとなり、注目を集めている。
本研究は、脱液胞化技術を用いて作成された、高い翻訳効率を持ち、生体内の翻訳機構を忠実に再現できる植物培養細胞由来の試験管内系を用いることにより以下の植物特有のRNAサイレンシング機構、特に小分子RNAを介した翻訳抑制機構の分子機序を、生化学的手法によって明らかとすることを目的とした。
平成24年度は、平成22、23年度に作成したタバコ及びアラビドプシス培養細胞由来の試験管内翻訳抑制を用いることにより以下の成果を得た。
1.miRNA標的配列の相補性、またその位置は植物RISCによる翻訳抑制に大きな影響を与える。
2.植物のRNA誘導サイレンシング複合体(RISC)は標的との結合に高い相補性を必要とする。
3.植物RISCが標的mRNAの5'UTR及び3'UTRに結合した場合、翻訳の開始段階を抑制する。
4.タンパク質コーディング領域に強く結合した植物RISCはリボソームの進行を阻害する。
これらの結果より植物RISCは結合する場所により異なるメカニズムで標的の遺伝子発現を制御していることが明らかとなった。
