Research Abstract

マンナン結合タンパク質MBPは、樹状細胞レクチンDC-SIGNと類似の糖結合特異性を示すことが知られている。申請者は、本年度において大腸がん組織におけるMBPリガンド糖鎖の特徴および発現パターンを調べ、臨床病理学的意義を検討した。申請者は、まず大腸がん組織におけるMBPリガンド糖鎖の発現パターンを詳細に調べた。MBPによる大腸がん組織の染色は、フコースに特異性をもつAleuria aurantia lectin(AAL)の存在下によって顕著に阻害されたことから、MBPはがん組織上のフコース残基を認識していることが明らかになった。さらに、抗ルイス抗体とMBPによる二重染色の結果から、MBPは大腸がん組織上のルイスB糖鎖[α-Fuc(1→2)-β-Gal-(1→3)-(α-Fuc-[1→4])-GlcNAc]を認識していることが示唆された。
興味深い事に、非癌領域上に発現するルイスB糖鎖はMBPによって染色されなかった。このことは、大腸癌組織上には従来の血液型関連ルイス糖鎖とは異なる"癌関連ルイス糖鎖"が存在することを示唆し、MBPはこの癌関連ルイス糖鎖のみを認識している可能性が考えられる。一方、非還元末端にシアル酸が付加されたシアリルルイスA(CAI9-9)の発現パターンは、MBPリガンドの発現とは著しく異なることも明らかにした。次に申請者は、大腸がん組織196例を用いてMBPリガンド糖鎖の発現を調べた。そのうち、77症例(39.3%)がMBPで染色され、119例では染色がみられなかった。一方、近傍の非癌領域67症例においてはいずれもMBPでの染色はみられなかった。BPによる染色の臨床病理学的分類を行ったところ、MBP染色の陽性率は60歳以上の大腸がん患者において有意に高く(P=0.0020)、分化度の低い癌に比べて高分化癌において有意に高い(P=0.0118)ことが明らかとなった。申請者の所属研究室ではこれまで、ヒト結腸がん細胞株SW1116細胞表面に、ルイスA糖鎖のタンデムリピート構造が存在することを報告している。本研究による成果は、ヒト大腸がん組織上において同様の癌関連ルイス糖鎖が存在することを示唆し、MBPが大腸がんの有効なマーカーとなりうることを示すものである。
