Research Abstract

世界の農業生産に甚大な被害を与えている根寄生雑草は植物の根から分泌される発芽刺激物質に曝されて初めて発芽する。代表的な発芽刺激物質としてはストリゴラクトン(SL)がある。このSLは、根圏ではアーバスキュラー菌根菌の菌糸分岐誘導物質として作用し、植物体内では植物ホルモンとしても作用する。本年度は、以下の項目を中心に調べた。
「養分条件がナタネの発芽刺激物質生産に与える影響」
これまでにナント大学(フランス)との共同研究により、アブラナ科植物の特徴的な二次代謝産物であるグルコシノレートから生成されるイソチオシアネート(ITC)、2-phenylethyl ITCがP. ramosaに対する主要な発芽刺激物質であることを明らかにしている。また、ナタネはAM菌の宿主植物と比べて分泌するSLは非常に微量ではあるが、既知SLのうちorobanchyl acetateを分泌していることを明らかにしている。そこで次に、養分条件がナタネの2-phenylethyl ITCおよびorobanchyl acetate分泌に与える影響を調べた。その結果、窒素欠乏と硫黄欠乏は2-henylethyl ITC分泌を減少させること、窒素欠乏とリン欠乏は若干orobanchyl acetate分泌を促進させることがわかった。
「新規Arabidopsis SL生合成変異体のSL解析」
オーストラリアとの共同研究により、新規Arabidopsis SL生合成lbo変異体、その野生株Ws-4およびSL生合成max1変異体、その野生株colombia-0の植物体に存在するSLの同定を試みた。その結果、SL生合成前駆体として報告されているcarlactoneが、lbo変異体およびmax1変異体の根からLC-MS/MSにより検出された。

Strategy for Future Research Activity

SL生合成前駆体として報告されているcarlactoneは、SLよりも不安定で壊れやすいため、これまでSLで行ってきた抽出方法および精製方法を改善させる必要がある。
