Research Abstract

多くの国において死亡者数の高い循環器系疾患の低侵襲的な治療法として、カテーテルやステントを用いた血管内治療の導入が増加している。しかしながら、医療デバイスの接触、留置による血球破壊や凝集など、血球細胞が受ける影響が問題となっている。本研究では、デバイスに対する力学的・生物学的応答の評価を可能とする、多様な膜強度を持つ血球モデルの開発をおこなってきた。血球モデルとして扱いが簡便である脂質二重膜小胞(リポソーム)、および赤血球膜に作用してナノサイズの孔を開ける膜孔形成タンパク質を利用して、膜の物性を硬化・脆弱化させた血球モデルの開発を目指した。昨年度は、膜孔タンパク質として、黄色ブドウ球菌Hlg、大腸菌FhuAΔC/Δ4L、およびヒラタケPleurotolysin (Ply)を使用し、反応させる脂質の電荷および形状を変化させることにより、膜孔の形成効率および電気生理学的性質が変化することを明らかとしてきた。
本年度は、平面脂質二重膜システムを使用してPlyの電気生理学的性質の解析を進め、Plyがスフィンゴミエリン(SM)脂質に特異的に作用し、膜孔が陽イオン選択性を有することを明らかとした。またCryo電子顕微鏡により、SMの脂肪酸側鎖の炭素数が18以上であることが膜孔の効率的な形成に必須であることを明らかとした。
さらに、異なる電荷および形状を有する平面脂質二重膜を作成し、電圧負荷時に膜に生じる微少な電流ノイズを計測し、膜安定性を解析した。その結果、逆コーン型脂質であるSMが存在することで、膜安定性が減少することが示唆された。昨年度の研究成果から、脂質と膜孔のコンビネーションを変化させることが、脂質二重膜強度を制御する重要因子であることが明らかにされていたが、本年度の実験結果から、膜を構成する脂質の種類を変化させることも、様々な膜強度を有する血球モデルの開発に重要な要素であることが示唆された。
