Research Abstract

これまで、細胞-細胞間接着分子であるE-カドヘリンのモデル分子の固定システムを用いて、細胞間接着の制御機構について検討してきた。前年までに肝細胞の接着とDNA合成能などによる分化維持について解析してきたが、今年度はマウスの胚性奇形腫細胞であるF9細胞を用いて未分化な細胞におけるE-カドヘリンの役割と、分化によるE-カドヘリンの機能的変化について検討した。
F9細胞はゼラチンやコラーゲン・ファイブロネクチンなどのインテグリン仲介型の細胞外マトリックス上ではコロニーを形成して増殖するのに対し、E-カドヘリンモデル分子上では活発な細胞移動が観察され、また細胞骨格の形成にも大きな変化が見られた。このE-カドヘリンモデル分子上での細胞移動は固定化したE-カドヘリンに依存したものであることが確認され、また分化した細胞である肝細胞や乳腺細胞では細胞移動がみられず、逆に細胞の凝集が観察された。これらのことから、分化の度合いによってE-カドヘリンの役割・制御機構が異なっていること、また細胞-細胞間の相互作用と細胞-細胞外マトリックス間の相互作用との協奏的な制御機構が細胞の分化に重要であることが示唆され、このモデルシステムを用いることで細胞接着と分化の更なる解析が行われることが期待された。
遊離型と固定型とでシグナルが異なることが示唆されている増殖因子のシグナル伝達系を解析するために、EGFモデル分子を用いた。EGF受容体を多数発現しているヒト扁平上皮細胞ガン細胞株であるA431細胞をEGFモデル分子上に播種すると、DNA合成能の減少、細胞増殖抑制がおこりまたEGFのシグナル系の下流にあるMAPK(ERK1/2)のリン酸化の持続が見られ、遊離型EGFのシグナル伝達とは異なる挙動を示すことも明らかにした。また、細胞骨格の形成の変化も確認され、EGF刺激の方法によって細胞に伝わるシグナルが異なり、細胞応答も異なることが示唆された。
