Research Abstract

種々の非天然型核酸が合成され、アンチセンスおよびアンチジーン核酸医薬としての応用が検討されている。その中でもペプチド核酸(PNA)はヌクレアーゼ活性が高くかつ天然型核酸と安定なハイブリッドを形成することで注目されている。PNAを細胞特異的送達しつつ、かつ標的核酸存在下で速やかにPNAをリリースする送達担体として、リガンドタンパク質とDNAからなるタンパク質・DNAコンジジュゲートを作成した。このコンジュゲート体は、肝細胞特異的リガンドタンパク質としてのアシアロフェツイン(AF)とPNA担持サイトとしてのDNAから構成されている。コンジュゲート体は血清存在下においてもPNAを安定に保持した。さらに、標的核酸と同様の塩基配列を持つDNAを加えることで、速やかにPNAを放出した。また肝実質細胞への取り込みをin vitroで解析した結果、PNA単独に比べコンジュゲート体を用いることで著しくPNAの取り込みが増大した。さらに取り込まれたPNAが細胞核に集積していることが見いだされた。PNAの細胞核への集積はインキュベーション開始後4時間頃から観察されるが、過剰量のフリーなAF存在下で抑制された。つまり、AFのレセプターであるアシアロ糖タンパク質レセプターによりコンジュゲートと共に内在化したPNAが、細胞核へ集積したと考えられる。内在化されたPNAのエンドソームからの放出および核への集積機構は不明であるが、細胞特異性と核への集積性を兼ね備えたキャリアとして興味深い。
