Research Abstract

本年度の研究では,好熱菌Thermus thermophilus由来リボソームタンパク質L5と5S rRNA断片との複合体の2.5Å分解能結晶構造および超高度好熱古細菌Pyrococcus horikoshii由来真核生物翻訳因子eIF-5Aホモログ(Pho IF-5A)の2.0Å分解能結晶構造を決定した.
我々は前年度の研究で,大サブユニットの結晶構造では一部決定されていない,リボソームタンパク質L5の完全な結晶構造を明らかにした.本年度は,より詳細な核酸タンパク質複合体の安定化とRNA-タンパク質認識に関する知見を得るため,L5と5S rRNA断片との複合体の結晶構造解析を行った.L5は保存された5S RNAループ-アミノ酸残基同士の水素結合による特異的で強固な相互作用により結合し,非結合型L5と比較してRNA結合による有意な構造変化は見られなかった.また,結晶格子中で隣接したL5は1分子のRNA断片を共有して結合し,本来23SrRNAと結合する湾曲したβシート表面において同様な結合様式で2重らせんRNA部分と非特異的に結合することも確認された.これらの2領域における5S rRNA-L5-23S rRNA相互作用はサブユニット構造における安定化を示唆している.
真核生物翻訳因子eIF-5Aは細胞の増殖と生存に関与し,機能的な修飾アミノ酸,ハイプシンを含むことが知られている.eIF-5Aは2つのドメイン(N-ドメイン,C-ドメイン)から構成される.本研究の好熱古細菌eIF-5A(Pho IF-5A)ホモログと,既報の二種の古細菌由来eIF-5Aホモログとを比較すると,ハイプシン修飾部位を含むN-ドメインはアミノ酸配列および立体構造ともによく一致しているが,それに対してC-ドメインの配列保存性は低く,立体構造は一致していなかった.さらにPho IF-5A結晶格子に含まれる独立した3分子を比較すると,同様にC-ドメインが異なる立体配座をとりうることが明らかとなった.このドメイン間の可動性を解析したところドメイン間リンカーの自由度は7°であった.分子表面に関して,N-ドメインは正電荷に,C-ドメインは負電荷に偏っていることから,各ドメインがタンパク質,核酸のいずれにも相互作用し,その機能に可動性が関与していることを示唆する.
