Research Abstract

本研究では、筋小胞体カルシウムATPaseの結晶構造解析により、膜蛋白質の結晶中の脂質二重膜の可視化を主な目標とした。これは、カルシウムATPaseの結晶化にあたって脂質を添加しているために脂質二重膜が形成されていることを利用するものである。脂質二重膜は低分解能領域にしか寄与しないので、普通の結晶解析では棄てている。また、重原子置換体による位相情報は、低分解能領域では信頼性が無い。このために、コントラスト変調法が試みられて来た。また.MAD法(多波調異常分散法)の適用も試みられるべきである。この目的のために、現在最も安定して作成できるカルシウムATPaseの結晶である、Mg/F複合体結晶を用い、庶糖と金グルコースによる密度変調を試みた。この結果,庶糖の方が可変できる範囲は広いものの,格子定数の変化も著しいことがわかった。金グルコースの場合,最大濃度は15%程度であったが,金による異常分散効果を利用できるため,コントラスト変調を用いる必要はない。また、格子定数の変化も無視できる程度であることがわかった。そこで,最大濃度の金グルコースを用いて,SPring-8でMADデータの収集を行った。その結果は現在解析中であるが、統計的には十分な信号が得られた。MADデータから位相を決定することは、重原子位置が離散的でないため無理であるが、蛋白質部分からの位相を改良することは十分可能であろうと考えられる。この部分の方法論はまだ確立していないので、今後の課題としたい。
