Research Abstract

F1ATPase(F1:サブユニット組成α3β3γε)は、呼吸鎖が形成する膜を隔てた水素イオンの濃度差をATPに変換するATP合成酵素の膜から突き出した部分である.私たちは回転触媒機構の詳細を知る目的で、好熱菌F1・α3β3γε複合体を用いて、異なるヌクレオチド結合モードがひきおこす構造変化の詳細を調べている.
今年度は、Mg-free ADPが1個だけβサブユニットに結合したF1の結晶構造と触媒機構との関連について議論した。Mgヌクレオチドを結合したβサブユニットが2個以上あるF1の従来の結晶構造との比較により、ヌクレオチド結合の変化に伴うα3β3コア部分の構造変化、γサブユニットの構造変化を特定することができた.ただしMg-freeの制約の十分な考慮が必要であった。
この構造をF1の従来の結晶構造と比べると、従来の結晶構造中で活性サブユニットと思われるclosed β DPサブユニット型の構造が見られない。これが通常のmuliti-site反応に比べてunisite反応が非常に遅いことと照合している。またγサブユニットを2個ヌクレオチドを結合したβサブユニットをもつF1構造のγサブユニットと比べると、約25度ねじれていた。これは1分子計測から得られた、産物ADP Piの放出に伴う30度回転と大きさ、方向ともに合致した。Mgが存在しないため好熱菌F1は軽く阻害された状態にある。これに対応してεサブユニットがα3β3コア部分にそのヘリックス部分を差し込んでいた。
