Research Abstract

α-カテニン遺伝子に変異を持ちそのためα-カテニンの発現が陰性である紳胞(DLD-1細胞)とこの細胞にα-カテニンの発現ベクターを導入してα-カテニンを発現させた細胞を用いて実験を行ってきた。この細胞はE-カドヘリン遺伝子は正常であるので、α-カテニンに依存したカドヘリンからのシグナルを調べるのに最適である。細胞の増殖性を詳細に調べた結果、シャーレに接着させた場合には両者の間で差がなかったが、シャーレへの接着を抑えてやると、α-カテニン陰性細胞に比べてα-カテニン発現細胞の増殖性は著しく低下していることが判明した。抗体でカドヘリンの機能を抑えると、増殖の抑制は見られなくなった。このことは、浮遊状態の細胞ではカドヘリンからα-カテニン依存性のジグナルが出ていることを示している。そこでこの現象に関与するα-カテニン機能領域の同定を行った。その結果、カドヘリン依存性のコンパクションを引き起こすのに必要な領域と一致することが判明した。さらに、Rbファミリーのp130のリン酸化状態を調べたところ、α-カテニン陰性細胞では接着状態と浮遊状態とで差がなくどちらの条件下でもリン酸化を受けていたが、α-カテニン発現細胞では接着状態から浮遊状態へと変えることによりリン酸化の程度が著しく低下することが判明した。Rbタンパク質のリン酸化は細胞周期の進行と密接に関わっており、カドヘリンからのシグナルがRbタンパク質のリン酸化を抑えたという結果は大変興味深い。
