Research Abstract

S対立遺伝子間の優劣性に関しては、花粉側でのSP11の発現に着目し高い席を行い、SP11の転写制御領域であると考えられるATGより-300bp領域では69.2-85.5%の相同性を有することがわかった。またこれらの領域にはDNAのメチル化の修飾を受けるとされるCpG,CpNpG配列がハプロタイプ間で保存されて存在している場所と、ハプロタイプ特異的な場所が存在していた。
昨年度、見出した一側性不和合性を制御する遺伝子座は、S遺伝子型の分離とは一致しないことを明らかにし、少なくともここで見出した一側性不和合性において、花粉側表現型を決める遺伝子座とS遺伝子座は、独立の遺伝子座によって制御されていることを明らかにした。今後は、柱頭側一側性不和合性の遺伝分析を行うとともに、花粉側を制御する遺伝子を単離するためのマッピングを行うことを計画している。
SLGが欠失していると考えられたB. campestris S36系統を使って、柱頭cDNAライブラリーを作製した。eSRK,SRKなどのカタログ化することを目的として、SLG45 cDNAをプローブとして、スクリーニングを行い、positive cloneについて、その両末端から塩基配列を決定した。独立な100クローンの部分塩基配列を決定したが、そのほとんどが、SLR1,SLR2であり、eSRK,SRKのカタログ化は困難であった。
Brassica campestris S9系統に0.2%EMS処理を行ったM2世代のミツバチスクリーニングから見いだした和合と考えられる7系統について、顕微鏡観察を行ったところ、1系統(#11-11)のみが和合性と判断できた。S9系統との相互交配実験から、この和合性が柱頭側での変異により和合性になったことが明らかになった。この和合性系統と他のS系統との交配を行い、遺伝解析を開始した
