Research Abstract

1。トポIIαおよびβをそれぞれ特異的なsiRNAによりノックダウンして、染色体の凝縮に対する影響を調べた。αの不在時には、βがある程度補完するが、両方ともなくなると、分裂後期において染色体の分離が押さえられることを示した。しかしながら、分裂期染色体の主要なスカッフォルド蛋白とされているトポIIαがほぼ完全に染色体から消失したにもかかわらず、染色体の凝集はほぼ完了していることがわかった。さらに、抗がん剤VP16、ICR193の作用を調べ、トポIIαがノックダウンされた場合には、低濃度の薬剤でも十分な効果が得られる可能性を示した。
2。Nap1に結合する蛋白Nbp2の機能を遺伝学的な解析により調べた。Nbp2欠質変異が温度感受性を示すことを手がかりとし、マルチコピーサプレッサー、およびデイリーションサプレッサーを得、その結果からNbp2はMapカイネースを通じて細胞壁の統合と、蛋白フォスファテースを介してミトコンドリアの分配という2つのプロセスに関わっていることを示した。
3。クロマチンリモデリング複合体の構成成分とされるRsc58似ついて、温度感受性変異株を作成し、それをもとにマルチコピーサプレッサーの解析、ツーハイブリッド解析、免疫沈降、レーザースキャンニング傾向顕微鏡を用いた細胞周期解析を行った。その結果Rsc58はRsc4や転写因子Swi6と相互作用をしていること、それを介してクロマチンの構造と転写活性とに影響していることが明らかになった。
