Research Abstract

(1)ヒトおよびマウスの培養細胞を用いて、細胞周期S期の進行に伴う複製の進行過程を、S期における複製タイミングと複製部位の核内配置、染色体バンド構造、さらに複製フォークの進行速度について解析し、これらが密接に関連することを明らかにした。特に、複製フォークの進行速度がS期の初期から中期にかけて一端減速し、後期に最も速くなることを、独自に開発したDNAファイバー上で複製フォークの進行速度を測定する方法により明らかにした。すなわち、染色体上でS期初期に複製するRバンド領域からS期後期に複製するGバンド領域の境界のR/G遷移領域、およびマウス細胞のセントロメアヘテロクロマチン周辺領域の複製フォークの進行速度か最も遅くなることを見出した。また、マウス・セントロメアヘテロクロマチン周辺領域の複製起点の活性化と複製フォークの進行速度はヒストンのアセチル化により制御されていることを明らかにした。さらに、カンプトテシン(CPT)処理によるDNA損傷時に作動するチェックポイント制御についても検討し、各種特異的阻害剤存在下での複製フォーク速度やDNA合成量を調べる実験から、DNA損傷時に、S期チェックポイント因子PI3KKが複製開始点の抑制と複製フォークの安定性に貢献することを見出した。
(2)複製・転写等の核内イベントへの核マトリックスの関与を明らかにするため、転写誘導される熱ショックタンパク質HSP70遺伝子領域の核マトリックスとの結合状態について転写ファクトリーを含めたモデルを提示すると共に、転写されないゲノム領域が、複製前に核マトリックスに結合し、複製後に直ちに離れることを明らかにした。
(3)分子コーミング技術を確立し、DNAファイバー上での複製解析へ応用する一方、インプリント遺伝子の複製がアリル間で非同調的なタイミングで複製することを独自の方法で証明した。
