Research Abstract

MT1-MMP遺伝子は、癌の進展に重要な役割を演ずることが示唆されている。一方、同遺伝子欠損マウスには重篤な骨化不全が生じ、コラーゲン基質への血管新生にも影響を与えることが示唆されている。このように、様々な細胞のコラーゲン関連基質内への浸潤性移動、およびそこでの増殖に関わると考えられる。本研究期間において我々は、MDCK細胞が癌遺伝子;Srcの導入によりコラーゲンゲル内への浸潤性を示すようになり、このときMT1-MMP発現上昇を伴うことを見出した。さらにMT1-MMP遺伝子プロモーター内にsrc結合領域を同定した。また、MT1-MMP遺伝子は、複数のホモオリゴマーとして細胞膜に局在することでMMP-2の活性化能を高め、ゲラチンへの浸潤性を高めることを報告した。また、このオリゴマー形成に与るヘモペキシン領域(HPX)を強発現させると、MMP-2の活性化が抑制されると共に、ゲラチン浸潤活性も阻害されることを報告した。6種類存在するMT-MMPのうち、MT4-MMPおよびMT6-MMPは、MT-MMPのうち特異な膜結合様式(GPIアンカー型)を示すことを報告した。またMT6-MMPは、血清内のクラスタリンと結合し、活性がマスクされた状態にあることが見出された。一方、MT5-MMPは、大脳、小脳、海馬を中心に神経系で特に高い発現を示すが、我々は、脊髄後根神経節(DRG)細胞を用いた神経軸索伸長のモデルにおいて、この軸索伸長はラミニンコート上で誘導され、脳特異的な基質であるヘパリン結合性プロテオグリカンの存在下では抑制され、さらにはこのプロテオグリカン上に外来性に活性型MT5-MMP(可溶型)を添加することで再び軸策伸長が再誘導されることを見出した。加えて、同ノックアウトマウスを用いたマウス坐骨神経結紮モデルにおいて、MT5-MMPが神経可塑性に関わることを報告した。
