Research Abstract

染色体複製モニターシステムには、DNAポリメラーゼ構成因子の動態変移が細胞周期制御のキーシグナルとして機能する。申請者らは、大腸菌の染色体複製サイクルの制御システムにおいて、DNA上にロードされたスライディングクランプ(DNAポリメラーゼIIIβサブユニット)が複製開始因子DnaA蛋白質を不活性化するというRIDA(DnaAの制御的不活化)機構を見出している(Cell,1998;EMBO J.,1999)。また、RIDAにはHdaという、我々が新規に見出したタンパク質が直接関与している(EMBO J.,2001)。このDnaA不活性化は重複複製開始を防止し、細胞周期における1回性複製を維持するのに必須な機構である。本計画研究では、RIDAの分子機構とその制御機構の解明、および、複製異常を感知して細胞分裂を阻害する新しい制御系の機構解明を日指すものである。研究実施計画に沿って、本年度は、まず、RIDA因子間の相互作用機構、および、部位特異的変異導入法による各因子の機能構造に関する生化学的解析を進めた。その結果、Hdaタシパク質のドメイン構造とDnaAタンパク質のATP結合/水解モチーフとをアミノ酸残基レベルで明らかにすることができた(投稿準備中)。さらに、RIDAに必要な、DNA-Hda-クランプ複合体の基本構造を明らかにできた(投稿中)。これらはRIDAの素反応解明のため、必須な情報を供給する。また、新規のクランプ結合性因子を生化学的に探索、同定した。さらに、RIDA欠損により過剰複製を起こす株、およびその抑制変異株のジーンチップ解析・プロテオーム解析などにより、チェックポイント制御に関わる可能性がある因子を探索し、約20種の候補因子を見出した。これらの機能解析を個別に進め、複製サイクル制御機構に直接関わるものを見出した(Genes Cells,2003,他、投稿中)。
