Research Abstract

セミインタクト細胞を用いたin vitro輸送系において、輸送反応中のGFP-importinβ(輸送担体)とGST-NLS-GFP(輸送基質)1分子の挙動を、アセンブルした核膜孔複合体上で直接捉えることに成功している。輝点の寿命と蛍光強度の定量的解析より、輸送反応中に分子が核膜孔複合体上に滞在する時間及び1つの核膜孔複合体がimportinβを結合しうる分子数とimportinβに対する結合定数を求めた結果、以下の知見を得た。1)核膜孔複合体上には、importinβに対して数pmolオーダーの極めて強い少数の結合サイトと、およそ70nMオーダーの100〜200の多数の結合サイトが存在する。2)importinβ単独(基質とRan非存在下)では、核膜孔複合体上に短く滞在するもの(約1s)と、長く滞在するもの(約6〜10s)が見られるので、上記1)の両結合サイトと相互作用すると考えられる。3)importinβ/基質複合体(Ran非存在下)は短く滞在するもの(約1s)のみが見られ、核膜孔複合体上の弱い結合サイトのみと相互作用すると考えられる。4)importinβ/基質複合体(Ran存在下)は短く滞在するもの(約1s)と長く滞在するもの(約3s)がみられたが、長い滞在時間はimportinβ単独の滞在時間よりも短かった。1分子解析と同条件下において、輸送担体と輸送基質が核に集積する速度を求めたところ、輸送担体と基質ともに、長い滞在時間と核内集積速度が一致することが判明した。このことから、輸送担体が核膜孔複合体上の強い結合サイトに相互作用することが通過反応に重要であると考えられる。また、Ranの影響で滞在時間の変化が見られることから、強い結合サイトの性質がRanの影響で変化すると予想される。これら一連の結果は、一分子可視化技術を応用して得られた最初の知見であり、研究成果は十分に得られたと思われる。
