Research Abstract

細胞の形態変化や移動は、発生において重要な役割を果たしている。しかし、その制御機構の生体における解析は遅れている。その主な理由として、生体内で細胞の形態変化・運動を経時的に観察し、そのメカニズムを解析する系が整っていないことが挙げられる。そこで本研究では、ショウジョウバエや哺乳動物の脳組織片を用いて、細胞形態・運動の制御機構を解析を進めた。
後藤らは、翻訳後修飾に注目して解析を行った。発生において糖修飾やプロセッシング等の翻訳後修飾は重要な役割を果たしている。しかし、そういった修飾がどのように制御されているのかについては、ほとんどわかっていない。翻訳後修飾が行われる場は、主としてゴルジ体である。そこで、様々なゴルジ体マーカーを用いて詳細な細胞内イメージングをおこなったところ、驚いたことに、ゴルジ体には多様な種類があることが明らかとなった。この結果が示すところは、我々が見いだした多様なゴルジ体によって、多様な翻訳後修飾が制御されている可能性である。そこで、遺伝学的、生化学的解析に加え、実際の糖修飾が行われている過程をイメージングしたところ、それぞれ異なるゴルジ体では異なる糖修飾がなされていることがわかった。
川崎、平田らは、神経細胞の移動に注目して解析を行った。マウス胚の終脳において腹側接線方向への新規の細胞移動を明らかにした後、その細胞移動の制御機構について解析を進めた。その結果、終脳背側の新皮質領域には、この細胞の腹側接線方向への移動を促進する何らかの作用が勾配を持って存在すること。終脳腹側の線状体原基の領域には、この細胞の侵入を阻害する何らかの機構が存在することなどを見いだし、これらの細胞が移動する具体的な様子をイメージング技術を用いて明らかにすることに成功した。
