Research Abstract

生体膜に強く結合して生理機能を発現する膜機能性生体分子の精密立体構造と運動性を自発磁場配向膜と固体NMRの手法を用いて解析する新しい方法の開発を行い、以下の成果を得た。
1)自発磁場配向膜の開発とその性質の基礎検討ハチ毒の主成分であるメリチンとリン脂質Dimyristoyl-phosphatidylcholine(DMPC)をモル比で1:10に混合して調製した脂質2分子膜はゲルー液晶相転移点(Tc)以上において膜面を磁揚に平行に向けて自発的に磁揚配向する性質を持つ事が明らかになった。さらに光学顕微鏡で2分子膜の形態を観測したところ、直径が50μmにも達する巨大小胞を形成することが分かった。これらの結果から、メリチン-DMPC2分子膜小胞は磁場中で楕円体を形成して、自発的に磁場配向する性質が明らかになった。オピオイドペプチドダイノルフィン-DMPC脂質膜系も楕円小胞を形成し同様に自発磁場配向の性質を示すことが明らかになった
2)自発磁場配向膜中生理活性ペプチドの精密立体構造決定メリチンーDMPC2分子膜が自発的に磁場配向する性質を利用して、メリチンの膜結合構造を解析することを試みた。カルボニル炭素を13Cで標識したメリチンをリン脂質膜に再構成し、磁揚配向状態におけるカルボニル炭素の異方的化学シフト相互作用の解析を行った。この結果、メリチンはα-helixを形成しており、このhelixは膜貫通構造を持ち、膜法線と一定の傾き角を保ちながら膜中に挿入していることが判明した。さらに、この状態のペプチドは膜中で10^5Hz以上の速度で向きを変える動的な構造であることが分かった。一方、膜結合ダイノルフィンは、N-末端のみがα-helixを形成して膜に挿入している構造が明らかになった。
