Research Abstract

A simple analytical instrument based on surface plasmon resonance(SPR) for the the rapid screening toxic organic substance in environment samples has been needed for field use.
First, since conventional matching oil, which is used as the light interface between a prism and an Au coated cover glass, is unsuitable for field use, we developed a new light interface membrane based on a plasticized PVC. The membrane performance was best in the ratio of 0.5 di-2-ethylhexyl phthalate to 0.5 g tritol phosphate in 0.2g PVC at 80ーC
Second, in order to enhance the sensitivity of SPR signal, the efficient immobilization of anti-IgG to Au surface was investigated. The large SPR signal was obtained in the immuno-reaction between IgG and anti-IgG using 10-Carboxyl 1-decanethiol as the treatment agent in the test with some kinds of thiols.
Consequently, the good calibration curve was obtained in the range 0.6〜3.O× 10-3 mM 2, 4-di-chlorophenol, which was similar to dioxin.
From these experimental results, it became clear that SPR chemical sensor for the toxic low molecule organic substance like environment hormones could be constructed by development of the much more efficient immobilization-technique of antibody on Au surface.
