Research Abstract

We speculated that there is a stem cell of myocardial cell in heart tissue. But in spite of our search for stem cell, no evidence of the existence of stem cell was derived. We thought that the stem cell in the heart might be provided from outside of heart tissue, such as bone marrow. So we examined wheather the conditioning stimulus which is resembled to the heart tissue can induce the bone marrow cell to differentiate to the myocardial cell.
Method: Bone marrow cells were cultured on the silicone sheet. And then the sheet was periodically stretched at 60 Hz. The change of cell figure was checked everyday. And the expression of N-cadherin was examined because N-cadherin is specifically up-regulated in the gap junction of cardiomyocytes.
Results: Cultured bone marrow cells stretched periodically changed its shape and axis. Cells formed the connection between cells and became one theread like cell line. At last these threads formed network of cells which is very resembled to heart tissue. Immunostaining by anti-N-cadherin staining showed that gap junctions were formed between cells.
Conclusions: Although we cannot fine any sigh of exisitence of stem cell of cardiomyocytes. We showed that the environment of heart tissue has strong effect on the differentiation of bone marrow cells homed into the heart.
