Research Abstract |
1) To investigate in a cross sectional study the concentrations of keratan sulfate (KS) in the direct aspirated synovial fluid (SF) in patients with TMJ disc dis-orders. Sixty-two joints of 55 female patients (mean 34.1, mode 29 years old) were diagnosed as an anterior displaced disc with (WR; 24 joints) or without reduction (WOR; 24 joints), and with osteoarthritis (OA; 14 joints) by MR imagery.KS concentrations was quantified by electrochemilumine-scence immunoassay (ECLIA) with a monoclonal antibody (1-20 5D4). In 62 joints (100%), KS in SF was detectable. The mode and ranges of KS concentrations in the WR, WOR and OA groups was 1779.4, 617.22 and 387.75, and 280.32-39495.5, 64-8740 and 126.48-3510.5 ng/ml., respectively. This difference among the groups was statistically significant (p<0.05). 2) To investigate in a cross sectional study the frequencies of the TMJ or the sample with detectable Interleukin 6 (IL-6) in the recovered samples during the intra capsular irrigation in patients with disc disorders. Fourteen joints of 14 female patients (ranged 16 to 60, mean 37.0 years, mode 26 years) were diagnosed as an anterior displaced disc without reduction by MR imagery. The recovered samples were obtained approximately 10 ml each in the twenty 14ml-test tubes with the time-series collection. Each sample was concentrated into approximately 2 ml with vacuum freeze dryer. The concentration of IL-6 was quantified by enzyme-linked immuno-sorbent assay (ELISA) kit with the avidin-biotin amplified, two step sandwich method. In 12 joints (85.7%) or 116 samples of all 282samples(41.1%), IL-6 was detectable. The frequencies of samples with detectable IL-6 in the joint each were ranged from 90.0% to 0% (mean 41.4%, median 40%). Three joints with over 85% detectable frequency showed the different clinical feature from the undetectable 2 joints.
