Research Abstract

The beta-amylase of barley is one of the important enzyme in the mashing process during beer production.
The termostability of beta-amylase was surveyed in about 6,700 lines of barley genetic resouces inorder to find new and useful mutant gene and to realize the geographical distribution of the beta-amylase genes. Most of the lines were categorized into the types A, B and C.The proportion of the types A, B and C was 52%, 22% and 26%, respectively. The thermostability types in southwest Asia consisted of Types A, B and C.Most of the East Asian lines were Type A.More than 95% of the Ethiopian barley was type C.
Besides types A, B and C, we found new thermostability types including a beta-amylase-less mutant and a highly thermostable mutant.
In addition to thermostability the beta-amylase isozyme type was surveyed using means of Isoelectric Focusins (IEF). Thermostability type A and C lines were grouped into IEF-I type, and lines of type B consisted of the IEF-I and II types in the preliminary test. The IEF isozymes of beta-amylase were then surveyed in about 1,550 lines of the thermostability type B lines. The IEF type-II was 85% and the rest was the IEF type I.The IFF type-I is distributed mainly in the northern Europe.
A QTL analysis using the Steptoe/Morex (type B II/ B I) doubled haploid lines revealed that a QTL existed around the Bmyl locus.
