Research Abstract |
Effect of estradiol treatment on the development of PRL and GH cells in the anterior pituitary of neonatal rats : First, the effect of anti-alpha fetoprotein antibody on the development of PRL and GH cells in the pituitary of neonate was determined. Furthermore, influences of estradiol (E2) and serum fractions of neonate in PRL cells development in vitro were estimated using immunocytochemistry and RT-PCR.Serum E2 concentration did not elevate by Anti-AFP treatment. However, anti-AFP tended to be increased in PRL cell % of the pituitary but not in GH cell %. E2 was dose-dependently stimulated the increase in PRL cell % but not in GH cell %. While serum fractions of neonate inhibited the development of PRL cell in the pituitary determined by immunocytochemistry, PRL gene expression was in accord with the result of protein expression. These results indicate that E2 stimulates the development of PRL cell in anterior pituitary of neonatal rats, the stimulation may be regulated by a factor(s) in neonatal serum. Moreover, it is possibility that the translation into PRL protein is regulated by a factor(s) in neonatal serum.
