Research Abstract |
1.The xanthine (X) /xanthine oxidase (XOD) system serves as an in vitro source of reactive oxygen species and apoptosis of lymphoblastoid cell lines (LCLs) was induced with X/XOD treatment. Among cytokines such as interleukin (IL)-6, IL-10 and interferon-gamma, only pretreatment with IL-6 gave LCLs the resistance to X/XOD-induced apoptosis. Furthermore, the LCLs repeatedly treated with X/XOD and cultured with or without IL-6 showed much more structural abnormalities of chromosomes than those without X/XOD treatment. Colony forming efficiency of X/XOD-treated LCLs with IL-6 was significantly higher than those without IL-6, and even relatively higher than LCLs without X/XOD treatment. 2.The cloning procedures of these LCLs on the soft agar plates were performed and 5 to 10 clones in each sublines were established. No mutations were detected in exon 5,6,7 and 8 of p53 gene of these clones by screening with PCR-SSCP methods. 3.Approximately twenty cases of malignant lymphoma of thyroid gland were collected and the following studies were performed : (1) Microsatellite instability (MSI) was detected in high-grade diffuse large B-cell lymphomas at relatively higher frequency, although low-grade MALToma did not show MSI. (2) Among several cytokines affecting B cell growth, only interleukin-7 showed the difference in expression between thyroid lymphoma and chronic lymphocytic thyroiditis by RT-PCR analysis. (3) By using the methylation-specific PCR, the methylation status of death-associated protein-kinase CpG island in its promoter region was examined. The frequency of methylation was higher in thyroid lymphoma cases than in clonic lymphocytic thyroiditis cases.
