Research Abstract |
To evaluate the chicken folliclular immunen system, GC formation and the role of T cells are studied. A single GC was isolated from the chikcen spleen, immunized 7 days or 14 days earlier with antigen by the intravenous route. Rearranged TCR genes were amplified using primers for Vβ1 and Jβ. Sequence analysis revealed only few patterns of CDR3. The number of patern in 7 days-GC were 7/19, 4/16, 12/22, 7/10 (mean 10.3±1.4 patern/22 clone ; 46. 8%). The number of patern in 14 days-GC was 2/39 (5.1%). 1) These findings indicate that there is restricted T cell clonality within a single GC following antigen stimulation, and that the focusing of T cell repertory was took placen during GC formation. 2) RAG mRNA expression in GC was also observed at 5 days-GC.3) Deletion circular DNA, pruduced at V-D recmobination was observed in thymus, spleen, Peyer's patch, difusely infiltrated area of BF and 8 days-GC.
