Research Abstract |
Using the yeast two hybrid system of HBx, a 2619 bp gene was isolated and had putative Initiation methionine residues at 15 bp and stop codon at 1375 bp (encoding 453 amino acids). We have named this protein, X associating factor-1 (XAF-1). We made the GST-HBx, XAF-1 andmade sure the each other binding with in vitro binding assay. A human nothern blotting identified mRNA expression present in various tissues ubiquitously especially smooth muscle, liver and heart. Similarity searches showed that XAF-1 exibits 57% gene sequence identity to Drosophila melanogaster tumor suppressor gene (Tid gene). On the same period, other group reported a new tumor suppressor gene named hTid1 binding with papiloma virus type 16 E7. To comparing XAF-1, it revealed hTid1 was a alternative splicing form of XAF-1, and both genes expressed in many hepatocellular carcinoma cell lines ubiquitously. To characterize XAF-1, we overexpressed XAF-1 in HepG2 cells and stimulated Anti-Fas Ab, adriamycine and revealed that XAF-1 accelerated the cell apoptosis in both experiments. We hyposised that HBx bindinding to XAF-1 change the function of XAF-1 and causing hepatocarcinogenesis.
